RIPA buffer has been used as a “gold standard” for total protein extraction for 20 years. In fact, it fails to extract true total protein.  Due to its method defect, RIPA produce biased data (Western Blot, IP, ELISA, etc.) caused by uneven protein loss and leads to wrong interpretations.  Recently, it has been reported by more and more researchers that the data from RIPA are questionable.

Our P502 is an ultimate solution which can extract ture total protein (full spectrum protein profile).

The total protein extracted by our P502 can be used in all Western Blot, IP and co-IP assay.

  • Protein profiles extracted by denaturing and native lysis buffer are not identical. For a given application one lysis buffer might be superior to the other. This kit provides both denaturing and native lysis buffers for users to test and select the best one for a specific application.
  • Protease inhibitors is not necessary prior to extraction. However if downstream application takes significant amounts of time or the protein extract will be stored for longer period of time, addition of protease inhibitors to lysis buffer is recommended.
  • For determination of protein concentration, BCA kit (Pierce) is recommended.
  • To study protein phosphorylation, phosphatase inhibitors (such as PhosStop from Roche) should be added to lysis buffer prior to use.
